A user in our lab was working optimizing a method and noticed that his 1H
pulse width was changing after running an experiment. To confirm his observation,
I ran a series of fast HSQC experiments on the same sample and ran pulsecal
before each. The results are below:
exp# P1, us
2 14.27
3 14.85
4 14.73
5 14.69
6 14.87
7 15.00
8 15.21
9 15.57
10 15.20
11 15.17
12 15.06
I checked the tuning between each experiment and the wobble curve did not m=
ove at all.
I followed up with simply running pulsecal several times in a row to see if
it is just normal variability in the algorithm, but it gave me a consistent
value +/- 0.01us each time.
These tests were performed with a 15N-labeled protein sample in a 5-mm
Shigemi tube on a 600-MHz Bruker Avance NEO w/ TCI cryoprobe.
Does anybody have suggestions what could cause this variability? Thanks
